Effect of Substrate Analogue Administration on ¥Ä^(5-3)-hydroxysteroid Dehydrogenase Activity in Adrenals of Rat

Abstract
Rats were used as experimental animals throughout this study.
One hundred male rats were divided into 5 groups
Group 1: Control
Group 2: Dehydroepiandrosterone injected
Group 3: 17¥â-Estradiol injected
Group 4: 17¥á-Pregnenolone injected
Group 5: Allopregnanolone injected
For the female rats the same dividing as male rats was done.
Each group was subdivided into four groups consisting of 5 animals to be 2, 8, 16, and 24 hours groups. Immediately after sacrificing the animals, the adrenals were taken out for the determination of its ¥Ä5-3¥â-hydroxysteroid dehydrogenase activity.
The activities were determined by means of Beckman spectrophotometer D. U. after Rubin et al. The results of this investigation were as follows:
1. In the adrenals of control male rat, ¥Ä5-3¥â-hydroxysteroid dehydrogenase activities determined with dehydroepiandrosterone, 17¥â-estradiol, 17¥á-pregnenolone, and allopregnanolone as its substrates were 15, 19, 3, and I ¥ìg product/min. /mg. N. respectively. The adrenal of control female rat was shown a little higher activity for the enzyme.
2. Both male and female dehydroepiandrosterone administered groups were shown an increase in ¥Ä5-3¥â-hydroxysteroid dehydrogenase activity during the experimental period of 24 hours.
3. Both male and female 17¥â-estradiol administered groups were shown a decrease in ¥Ä5-3¥â-hydroxysteroid dehydrogenase activity during the experimental period of 24 hours.
4. The trend of variation of ¥Ä5-3¥â-hydroxysteroid dehydrogenase activities in adrenals of both male and female 17¥á-pregnenolone administered groups is similar to that of dehydroepiandrosterone administered those groups.
5. ¥Ä5-3¥â-hydroxysteroid dehydrogenase activities in both male and female allopregnanolone administered groups were increased at 2 hours after injection as much as 2.6 times control group, and decreased suddenly thereafter.
